Design and synthesis of an RNA interference kit

siRNAは、標的遺伝子のノックダウンツールとして広く使用されている。一方で、siRNAの配列デザインは、その目的に応じたデザインが必須で有り、例えば、核酸医薬などは、最適の配列デザインが創薬には必須であると考えられている。本支援では、依頼者からの研究目的に合わせて、最適なsiRNAをデザインし、研究の支援を行う。必要に応じて、siRNAを用いた解析支援も行う。

マイクロRNAは、セントラルドグマにおけるタンパク質合成の過程において、翻訳や転写を行う非コードRNAの一つである。siRNAが、ピンポイントに標的遺伝子をノックダウンするのに対して、マイクロRNAは、100以上のmRNAを標的とすることができ、それらの遺伝子が関わるネットワークのファインチューナーとして機能している(図1)。本支援では、これまでに主要な機能が判明しているマイクロRNAに絞って、それらのマイクロRNAを用いた機能解析のサポートなるマイクロRNAキットの配付を行う。

siRNA is widely used to knockdown target genes. In this program, we will design siRNA sequences according to the research purpose of the user and provide three types of siRNAs against one target gene.

We will also provide a microRNA (miRNA) library to the user. While siRNA only suppresses expression of a single target, one miRNA regulates the expression of multiple target genes. More importantly, aberrant expression of miRNAs is associated with the onset and progression of disease. Therefore, this miRNA library is useful for understanding the functions of miRNAs in a context relevant to the user.

マイクロRNA機能的スクリーニングの支援 【共同研究ベース】

① 配布キットを用いた機能的スクリーニング
配布キット(A~C)を用いた研究のデザイン、網羅的解析の実施などのサポートを共同研究ベースで実施可能である。

② 網羅的なマイクロRNAの機能的スクリーニング
多数のマイクロRNA(2000種類以上)を用いたより高度なスクリーニング希望の研究者に対しては、配布キットにはないマイクロRNAを用いた網羅的スクリーニングのデザイン、スクリーニング支援も共同研究ベースで実施可能である(ライブラリーの配布は行いません)。

※尚、共同研究ベース支援は、あらかじめ担当者(田原栄俊)に必ずお問合せ下さい。

【マイクロRNAライブラリーの使用例】

① 細胞増殖を指標としたスクリーニング

② 細胞の特定タンパク質の発現を抑制するマイクロRNAのスクリーニング

③ 細胞内の特定タンパク質の局在を変化させるマイクロRNAのスクリーニング

例えば、①~③のような網羅的スクリーニングの場合、下図のようなアッセイ系により、目的の機能性マイクロRNAを同定することができる。

【RNA干渉キット配付の条件及び注意事項】
Conditions and notes

上記以外に、RNA干渉キット(以下キット)の分与希望者は、キットの分与希望者が下記条件を遵守することを前提と致します。
In addition, please note the following conditions:

  • キットは、原則、科学研究費補助金を受給している日本国内の研究者(研究代表者または研究分担者)にのみ分与される。
  • キットの使用は、分与を受けた研究者のみとし、二次譲渡は禁止する。
  • キットは着払いによる配送とする。
  • キットの使用による研究成果は、すべて使用した研究者に帰属する。
  • キット使用者は、キット受領後半年以内に「RNA干渉キットユーザーレポート」フォームを用いて使用結果を報告する。 ただし、秘密事項保持等の事情がある場合には当面の期間はこれを要求しない。ただし、その期間は原則として受け付け後最長2年以内とする。
  • キット使用結果の公表の際には、使用者は当班に謝意を表さなければならない。
    また、公表した旨を当班に報告する。また、公表した旨を支援班に報告する。 (注)
  • 申込書に不備のある場合や上記注意事項が守られない場合は、受付出来ないことがある。
  • キットの使用によってキット使用者に何らかの不利益が生じたとしても当班はその責任を一切負わないものとする。
  • This program is only available to researchers with a Grant-in-Aid for Scientific Research (KAKENHI).
  • Transfer of siRNAs and miRNA libraries to other parties is prohibited.
  • Users must pay the delivery fee for siRNAs and miRNA libraries.
  • All research results generated using siRNAs or miRNA libraries belong to the user.
  • Users must report their research results using the “User Report Form” within 6 months of receiving the siRNAs or miRNA libraries. If there are confidentiality issues, users can postpone their reports for a maximum of 2 years.
  • Users must acknowledge our program when they report their results at academic conferences or in publications (please contact us prior to publication or presentation using the following e-mail address: cads@jfcr.or.jp).
